Traditionally RCA (replication competent adenovirus) is measured in permissive cells by a plaque-forming unit (PFU) assay which takes 10-14 days. Our Rapid RCA Assay Kit uses an immunocytochemistry staining protocol that requires only a two day incubation.
Figure 1: Assay Principle for the Rapid RCA Assay.
Figure 2: Staining Results from the Rapid RCA Assay. Serial 10-fold dilutions of Ad5 (2 x 10^12 viral particles/mL) were used to infect A549 cells in a 6-well plate for 48 hours. Anti-Hexon immunostaining was performed according to the assay protocol.
