From production to infection: measuring viral vectors across the workflow

Viral gene delivery using adenoviral, lentiviral and retroviral vectors typically follows a defined workflow:

  1. Packaging: introducing vector plasmids containing the gene of interest into packaging cells
  2. Physical titration: quantifying total viral particle concentration
  3. Purification: concentrating viral particles and removing contaminants
  4. Functional titration: measuring particles capable of infecting cells
  5. Transduction: infecting target cells with the virus, enabling delivery of the gene of interest

Because successful gene transfer depends on both viral quantity and infectivity, measuring viral preparations using both physical and functional titration is essential for reliable results.

Accordingly, our viral quantification products support vector workflows by enabling accurate measurement, functional assessment and monitoring across these stages.

Lentivirus quantitation

Traditional lentiviral vectors are derived from HIV-1 and efficiently deliver target genes into both proliferating and non-proliferating cells. Because each viral particle contains the p24 capsid protein, measuring virus-associated p24 during physical titration provides a rapid estimate of total lentiviral particle yield, supporting dosing calculations and gene delivery applications.

QuickTiter™ Lentivirus-Associated p24 ELISA

For highly accurate quantification of lentiviral physical titer via detection of virus-associated p24, providing a reliable estimate of lentiviral particle production.

Proprietary technology isolates virus-associated p24 from free p24, minimizing the overestimation of lentivirus titer, ensuring accurate assessment of particle yield.

Compatible with standard microplate readers, enabling straightforward integration into existing laboratory workflows without specialized instrumentation.

Includes an HIV-1 p24 standard, supporting quantitative analysis and consistent comparison of viral production across samples and experiments.

Adenoviral quantitation

Adenoviral vectors are widely used for gene delivery due to their high infection efficiency, strong transgene expression and ability to transduce most mammalian cell types. Following purification, adenoviral preparations can be quantified using a functional titer, which measures the proportion of viral particles capable of infecting cells.

QuickTiter™ Adenovirus Functional Titer Immunoassay Kit

Quantifies infectious adenovirus by detecting infected cells via immunocytochemistry, delivering biologically relevant titers for accurate dosing and reliable gene delivery experiments.

Functional immunocytochemistry readout of infected cells, providing a direct measure of infectious adenovirus rather than total particles for biologically meaningful titers.

More accurate than traditional plaque-forming unit (PFU) assays, reducing variability associated with manual plaque counting and improving confidence in dose determination.

Rapid turnaround (~2–3 days vs. ~10 days for PFU assays), enabling faster optimization of gene delivery experiments and shorter development cycles.

No agar overlay required, reducing hands-on time, and minimizing technical variability compared with plaque assays.

Recognizes all 41 adenovirus serotypes, supporting broad applicability across adenoviral systems without assay redesign.

Compatible with adenovirus amplified in HEK 293 cells, enabling straightforward integration into workflows.

Retroviral quantitation

Traditional retroviral vectors are based on murine leukaemia virus (MuLV), which are used to achieve stable integration of genetic material into host cell genomes. Each viral particle contains the p30 capsid protein, making detection of p30 a convenient method for identifying and quantifying MuLV-derived viral particles.

During physical titration, measuring the retroviral capsid p30 protein provides a rapid estimate of total vector yield, supporting dosing and gene delivery applications.

QuickTiter™ MuLV Core Antigen ELISA Kit

For accurate quantification of MuLV physical titers through detection of p30, providing a reliable estimate of retroviral particle production during quality control and batch release.

High analytical sensitivity (detects MuLV p30 down to 300 pg/mL), enabling reliable detection of low-level retroviral production  when vector yields are low.

Direct measurement of the MuLV p30 core antigen, providing a practical surrogate for viral particle abundance to support monitoring of retroviral vector yield.

Quantitation on a standard microplate reader, allowing seamless integration into existing laboratory workflows without specialized equipment.

Includes a recombinant MuLV p30 standard, enabling true quantitative analysis and consistent comparison of retroviral levels across experiments.
